Is there any way I can connect to udp port 53 through soft ether?

what you mean with through soft ether?
if you mean you will connect to the Server only tcp is possible.
and you should look if no other application have an listener on this port.
you can use netstate for this.
if you use an hub, all traffic will be bridged!

well I can not connect to a server with udp port 53

SoftEther VPN can listen on UDP port only for OpenVPN.
If you want so, please add 53 port to listener.
